热带顽拗性植物遗传转化与基因编辑技术研究进展

Research progress on genetic transformation and gene editing technologies for tropical recalcitrant plants

  • 摘要: 热带顽拗性植物是指在离体培养与遗传转化中表现出显著困难的热带植物,其育种改良受限于再生效率低下及基因型依赖性强等生物学特性。近年来,以CRISPR/Cas为代表的基因组编辑技术与多元化遗传转化体系的协同发展,为突破这一瓶颈开辟了新途径。文章系统梳理了热带顽拗性植物的生理特性与再生障碍的生物学基础,深入剖析了细胞全能性表达受限、防御反应激活及分子调控网络失衡等内在机制。重点综述了依赖组织培养和非组培依赖的原位转化体系两类关键转化策略的最新进展。同时,文章总结了基因编辑递送系统的演化趋势。最后,展望了热带顽拗性植物分子育种的发展前景,提出未来研究应立足于物种特有的生物学规律,采取针对性的技术策略,并注重多种技术路线的协同应用。

     

    Abstract: Tropical recalcitrant plants refer to tropical plants that exhibit significant resistance to in vitro culture and genetic transformation, with their breeding improvement being limited by low regeneration efficiency and strong genotype dependence. Recent advances in CRISPR/Cas-based genome editing and diversified genetic transformation systems, have begun to overcome these long-standing barriers. This article systematically reviews the physiological characteristics and biological basis of regeneration obstacles in tropical recalcitrant plants, and deeply analyzes the internal mechanisms such as the restricted expression of cell totipotency, the activation of defense responses, and the imbalance of molecular regulatory networks. This paper mainly reviews the latest progress of two key transformation strategies, tissue culture-dependent and in situ transformation systems without tissue culture. Meanwhile, the article summarizes the evolution trend of gene editing delivery systems. Finally, this paper discusses the development prospects of molecular breeding of tropical recalcitrant plants, and proposes that future research should be based on the species-specific biological laws, adopt targeted technical strategies, and pay attention to the coordinated application of multiple technical routes.
