大花序桉组培增殖苗衰退规律及复幼研究

Decline pattern and rejuvenation of proliferating Eucalyptus cloeziana plantlets in tissue culture

  • 摘要: 为明确大花序桉组培增殖苗连续继代的衰退特征,建立高效复幼技术体系,以10个川林系列无性系为材料,系统监测继代过程中增殖系数、芽苗形态及异常性状的动态变化,并以衰退典型无性系 “川林 73-86” 为模型优化复幼与生根配方。结果表明:不同无性系增殖衰退代数存在差异,但规律一致,连续继代15代后均出现增殖能力显著下降;“川林73-86”最优复幼培养基为 WPM+0.2 mg/L 6-BA+0.2 mg/L NAA+0.2 mg/L GA3+30 g/L 蔗糖,平均增殖系数达3.17;复幼芽苗生根后根系根毛浓密、韧性强,幼苗生长旺盛,苗木质量合格。揭示的衰退规律及构建的复幼技术,为大花序桉组培苗规模化稳定生产提供了关键技术支撑。

     

    Abstract: To clarify the decline characteristics of tissue culture proliferative seedlings of Eucalyptus cloeziana during continuous subculture and establish an efficient rejuvenation technology system, 10 Chuanlin series clones were selected as materials in this study. The dynamic changes of multiplication coefficient, seedling morphology and abnormal traits during the subculture process were systematically monitored, and the formula of rejuvenation and rooting was optimized with the typical decling clone "Chuanlin 73-86" as the model. The results showed that although there were differences in the number of proliferation decline generations among different clones, the change law was consistent: the proliferation ability significantly decreased after 15 consecutive subcultures. For "Chuanlin 73-86", the optimal rejuvenation medium was WPM + 0.2 mg/L 6-BA + 0.2 mg/L NAA + 0.2 mg/L GA3 + 30 g/L sucrose, with an average multiplication coefficient of 3.17. After rooting, the rejuvenated seedlings had roots with dense root hairs and strong toughness; the seedlings grew vigorously, and the seedling quality met the standard. The decline law revealed and the rejuvenation technology established in this study provide key technical support for the large-scale and stable production of Eucalyptus cloeziana tissue culture seedlings.
